To carry out the assay with a Saliva Alcohol Test Strip, Saliva should be collected according to the instructions given in the Instruction For Use included in the test kit. After collecting, it is preferable to perform the testing as early as possible; if not viable, the specimen should be stored properly. A general introduction about the collection, and storage of the specimens is given below.
Nothing should be placed into the mouth of the subject for at least 10 minutes prior to saliva collection, including food, drink, tobacco products or other materials.
Saliva specimen can be collected in a sputum cup or a clean container, or directly applied to the reaction pad of the test strip.
After 10 seconds, shake off the excess saliva.
Saliva Storage Condition
The saliva specimens should be tested as soon as possible after collection.
Color Changes under Enzyme Reaction method is used in this Saliva Alcohol Test Strip to detect the Saliva Alcohol in human Saliva specifically. The introduction of this Color Changes under Enzyme Reaction testing principle and reagent composition is given blow.
Rapid Alcohol Test is based on the high specify of alcohol oxidase (ALOx) for ethyl alcohol in the presence of peroxidase and enzyme substrate such as tetramethylbenzidine (TMB). The distinct color on reactive pad could be observed in less than 20 seconds after the tip was contacted with saliva samples with the ethyl alcohol concentration greater than 0.02%.
It should be noted that other alcohols such as methyl, propanyl and allyl alcohol would develop the similar color on the reactive pad. Fortunately to the saliva alcohol testing, these alcohols are not normally present in saliva.
Reagent Composition of Alcohol Saliva test strip: Tetramethylbenzidine (TMB) 0.12mg; Alcohol oxidase (EC) 0.5 IU; Peroxidase(EC) 0.35 IU; Proteins 0.15mg.
